Particle counters for continuous particle measurement in GMP cleanrooms

The adherence to strict cleanroom conditions is of great importance in numerous industries, especially in the pharmaceutical, biotechnology, and medical technology sectors. Good Manufacturing Practice (GMP) requires continuous monitoring of particle concentration in the air to ensure product quality and patient safety. Modern particle counters typically operate based on the principle of light scattering. An air stream is directed through a measurement chamber where particles are detected by a laser beam. The light scattering is registered by sensors and converted into information about the size and number of particles. Additionally, modern particle counters offer multi-channel analysis for simultaneous measurement of different particle sizes (e.g., ≥0.5 µm, ≥5 µm), integration into cleanroom monitoring systems, GMP-compliant data storage, and alarm/report functions.

Particle counters are used in all GMP classes (A to D), with monitoring requirements increasing with the cleanliness class. Zone A is particularly critical, where sterile work, such as aseptic filling, takes place. Continuous measurements are necessary here to detect even the smallest deviations in real time.

In Zone B and background zones (C and D), a combination of continuous and spot monitoring is usually used to ensure environmental control.

When selecting a suitable particle counter for GMP cleanrooms, the following aspects should be considered:

– Measurement accuracy and calibration: The devices must comply with ISO 21501-4 requirements and be calibrated regularly. The particle sizes to be detected must be defined in advance.
– Integration into existing monitoring systems: Particle counters should seamlessly integrate into a comprehensive cleanroom monitoring system, including pressure, temperature, and humidity measurements.
– User-friendliness and maintenance: Easy operation and low maintenance requirements reduce operating costs and increase reliability.
– Compliance features: Compliance with regulatory requirements such as FDA 21 CFR Part 11 and EU-GMP Annex 1 is indispensable.
